Anti-Idiotypic Antibody for PK Assay
Anti-idiotype antibodies are widely used in therapeutic antibody drug discovery because they specifically recognize and bind to the unique site (Idiotype) of the variable region of the antibody to be tested. Due to the similarity between anti-idiotype antibodies and anti-drug antibodies (ADA), they can be used as important references for immunogenicity and neutralization antibody studies. In addition, in the pharmacokinetic (PK) and pharmacodynamic (PD) analyses of antibody drugs, anti-unique antibodies can be used to detect the levels of antibody drugs (free, bound and total) in the blood.
Application of Anti-idiotypic Antibody in PK Assay
Anti-idiotype antibodies represent a class of reagents that are potentially optimal for analyzing the PK of fully human antibodies that have been developed as therapeutic candidates. Indeed, anti-idiotype antibodies are usually combined with ELISA to detect the concentration of target antibodies. Compared with drug target capture, anti-idiotypic antibody bridging reduced the false positive rate significantly due to the high specificity of reagents.
