A High-throughput Screening Method for Fast Identifification of sdAbs

Yeast surface display technology is a powerful tool for screening large libraries of proteins or peptides. In this technique, the protein or peptide of interest is fused to the surface of a yeast cell. By coupling this technique with flow cytometry and NGS, millions of sdAb clones can be sequenced in a single experiment, enabling the rapid identification of sdAbs against a target antigen.

The NBbiolab yeast surface display platform utilizes the "a-agglutinin" display system, where nanobodies are displayed at the N-terminus of Aga2p, fused with an HA tag at the C-terminus of the Aga2p subunit. The resulting fusion protein is secreted and anchored to the cell wall allowing nanobodies to be displayed on the cell surface thereby enabling their accessibility to soluble ligands.

Differences Between Phage Dispaly and Yeast Surface Display

Antibody display libraries are traditionally analyzed by isolating a limited number of clones in combination

with Sanger sequencing, which may not detect all desirable clones due to dominance of certain clones and

random selecrion. By contrast, yeast surface display combined with next-generation sequencing (NGS)

allows for far-greater insights into library diversity enabling the identifification of high-affifinity and specifificity antibodies against a target antigen.
